    为了探究重组酱油曲霉碱性蛋白酶(rAp)水解大豆分离蛋白获得的大豆肽对小鼠免疫功能及抗氧化能力的影响。将诱导表达后分离纯化的rAp对大豆分离蛋白进行水解,然后将水解液灌胃小白鼠,测定其水解液对小鼠免疫器官指数、小鼠血清溶菌酶活性和小鼠血清抗菌活力等小鼠免疫功能的影响;并通过测定谷胱甘肽过氧化物酶(GSH-PX)和过氧化氢酶(CAT)的活性,以及丙二醛(MDA)含量等进行小鼠体内抗氧化试验。进一步检测对小鼠血液中谷胱甘肽过氧化物酶(GSH-PX)、过氧化氢酶(CAT)活性和丙二醛(MDA)含量的影响。结果表明,大豆肽对小鼠免疫器官指数没有显著影响,但具有显著地提高小鼠血清溶菌酶活性和小鼠血清抗菌活力;对小鼠血清中的GSH-PX的活力影响不显著,但能显著提高血清中的CAT活力和显著降低MDA含量。由此可见,rAp水解大豆分离蛋白获得的大豆活性肽一定程度上能增强小鼠机体的免疫功能和抗氧化活性,在保健品行业为大豆的高值化利用提供了实验依据。

    Abstract:

    To explore the effects of soybean peptides from soybean protein isolate(SPI) hydrolyzed by the recombinant Aspergillus sojae alkaline protease (rAp) on immunity function and antioxidation in mice. In order to produce the rAp, the recombinant P. pastoris strain was constructed and inoculated in shake flask. SPI was hydrolyzed by the rAp, and then the soybean peptides were obtained. Furthermore, the immunity functions of the soybean peptides was to investigate the effects of the soybean peptides on immune organ indices, serum lysozyme activity and the anti-bacterial activity. And the antioxidation of soybean peptides was to determine the level of malondialdehyde(MDA), the activities of glutathione peroxidase (GSH-Px) and catalase(CAT). The results indicated that the immune organ indices had no remarkable change, but the serum lysozyme activity and the anti-bacterial activity were enhanced. In addition, the content of MDA was significantly lower to the control group. The activities of GSH-Px and CAT were significantly higher to the control group. In conclusion, the soybean peptides can enhance the immune function and anti-oxidative activities in mice.
