In order to provide scientific basis for the development and UV of food resources rich in chlorogenic acid, the protective effect and mechanism of chlorogenic acid on HaCaT cells injured by UVB were studied. MTT method was used to establish cell UV damage model and chlorogenic acid treatment method. The UV absorption of chlorogenic acid was detected by enzyme labeling instrument. The apoptosis level of cells was detected by flow cytometry. The free radical scavenging ability of chlorogenic acid was detected by ABTS method. The results showed that the radiation dose of UVB was 21.6mJ/cm2, and the concentration of chlorogenic acid was 100 and 150 μmol/L After pretreatment for 1 h, the cells were cultured for 12 h; Chlorogenic acid has strong absorption in UVB band (280nm-320nm); 100 and 150 μmol/L chlorogenic acid both significantly reduced UVB induced apoptosis; 150 μmol/L chlorogenic acid could effectively increase the ABTS+ free radical scavenging ability of UVB treated cells by 53%. Therefore, chlorogenic acid has strong anti skin photoaging activity in HaCaT photoaging model induced by UVB, and its mechanism may be related to the UV absorption and antioxidant capacity of chlorogenic acid.
