      采用响应面法对卵蛋白源胰脂肪酶抑制肽(PLIPA)的制备工艺进行了研究,经模拟胃肠道消化研究PLIPA的稳定性。C57BL/6J小鼠饲喂高脂饲料40 d后成功建立肥胖模型,灌胃PLIPA 28 d后处死,检测相关指标,研究PLIPA对营养型肥胖小鼠体质量和体形的控制效果。结果表明,卵蛋白浓度80 g/L,加入2240 U/g的碱性蛋白酶,在pH9.0,55℃水解4 h,制备的PLIPA对胰脂肪酶的抑制率最高,为56.06 ± 2.64%。经模拟胃液和胃液+肠液消化后,每毫克PLIPA对胰脂肪酶的抑制率分别为0.53%和0.46%,整体抑制率仍较高,稳定性较好。小鼠试验表明PLIPA复合组的体质量和Lee's指数显著低于模型组(P < 0.05),处理组的腹部脂肪系数、甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白(LDL-C)与高密度脂蛋白(H-DLC)等指标均低于模型组,表明PLIPA对于肥胖小鼠具有体质量减轻和降脂作用,其中PLIPA复合组控制体质量和体形效果最好。

      The preparation of pancreatic lipase-inhibiting peptide from albumin (PLIPA) was studied by response surface method, the stability of PLIPA was studied by simulated gastrointestinal digestion. C57BL/6J mice obesity model was successfully established after feeding high fat diet for 40 days, After 28 days of administration of PLIPA, the mice were killed and the related indexes were detected to study the control effect of PLIPA on body weight and body shape of obese mice. The results showed that when the concentration of albumin was 80 g/L, albumin was hydrolyzed by 2240 U/g alkaline protease at 55℃, pH9.0 for 4 h, the inhibition rate of PLIPA was highest (56.06 ± 2.64%). After digestion of simulated gastric fluid and simulated gastric and intestinal fluid, the inhibitory rate of per milligram PLIPA on pancreatic lipase were 0.53% and 0.46%, respectively, and the overall inhibition rate was still high and the stability was good. the body weight and Lee's index of the PLIPA complex group were significantly lower than that of the model group (P < 0.05). The abdominal fat coefficient, triglycerides (TG), total cholesterol (TC), and low-density lipoprotein (LDL-C) and high-density lipoprotein (H-DLC) of treatment groups were lower than that of the model group, which indicated that PLIPA had an effect of reducing body weight and fat in obese mice, and the effect of controlling body weight and body shape was the best in PLIPA complex group.
