      观察茯砖茶对非酒精性脂肪肝治疗效果及对肝脂合成和氧化应激的影响。选取2月龄雄性APOE-/-小鼠55只,连续高脂饲料喂养13周后随机抽取5只行HE染色判断成模率。造模成功后随机分为模型组,阿托伐他汀组,茯砖茶高、中、低剂量组,持续高脂饲料喂养并灌胃干预8周。同时选2月龄C57小鼠作为空白对照组,予以普通饲料喂养13周后予以生理盐水连续灌胃8周。实验末取小鼠肝脏组织用于肝功能、氧化应激、HE染色及RT-qPCR。病理学显示茯砖茶高、中剂量组小鼠肝脂变轻于模型组。与空白对照组比较,模型组肝组织GSH-PX、SOD活力较低,MDA含量、ALT活力、AST活力、NAS积分、SCD-1、PPAR-γ、HMGCR基因表达较高(P<0.05)。与模型组比较,茯砖茶不同剂量组肝组织MDA含量,ALT、AST活力,NAS积分,SCD-1、PPAR-γ、HMGCR基因表达较低,GSH-PX、SOD活力较高。茯砖茶通过抑制肝脂合成和减少肝脏氧化应激损伤而达到逆转早期NAFLD的效果。

      To observe the effect of Fu brick tea on lipid synthesis and oxidative stress injury in APOE-/- mice with non-alcoholic fatty liver. Fifty-five male APOE-/- mice(2 months)were continuously fed with high fat diet for 13 week, five of them were randomly selected for HE staining to estimate this model. The remaining fifty APOE-/- mice were randomly divided into model group, atorvastatin group and high, medium and low doses groups of Fu brick tea after successful modeling, all of APOE-/- mice were respectively intervened with normal saline or corresponding drug through gavage for 8 weeks. At the same time, C57 mice(2 months)were selected as the control group and fed with normal saline for 13 weeks, then given saline for 8 weeks. At the end of the experiment, liver tissues of mice were taken for liver function, oxidative stress, HE staining and RT-qPCR. Pathological morphology showed that the degree of liver steatosis in Fu brick tea of high and medium dose groups were lighter than model group. Compared with the normal control group, the activity of GSH-PX and SOD in the liver of the model group decreased; the content of MDA, the activity of ALT and AST, NAS score, the gene expression of SCD-1、PPAR-γ and HMGCR increased (P<0.05). Compared with the model group, the GSH-PX and SOD activity of Fu brick tea in different dose groups increased, the content of MDA, the activity of ALT and AST, NAS score, the gene expression of SCD-1、PPAR-γ and HMGCR were decreased (P<0.05). Fu brick tea can reverse the early NAFLD by inhibiting liver lipid synthesis and reducing liver oxidative stress damage.
