    摘要:

    目的 旨在选育一株乙醇耐受性优良且适合麦芽醋酿造的醋酸菌,同时提高其原料利用率及醋酸产量,从而改善成品醋品质。方法 比较了热带醋杆菌J-22(Acetobacter tropicalis J-22)、糖精葡糖醋杆菌J-15(Gluconacetobacter saccharivorans J-15)和沪酿1.01(Acetobacter pasteurianus HN 1.01)在不同乙醇体积分数(6%~10%)啤酒中的发酵能力,以产酸量(以乙酸质量浓度计)、乙醇转化率及感官评价为指标,确定综合评价最高的热带醋杆菌J-22为出发菌株,通过常压室温等离子体(atmospheric and room temperature plasma,ARTP)诱变结合高通量筛选(high-throughput screening,HTS)技术,最终选育出1株热带醋杆菌J-2276。通过测定在不同乙醇体积分数下诱变菌株热带醋杆菌J-2276和出发菌株热带醋杆菌J-22的生长曲线和麦芽醋发酵性能,进一步验证了该诱变菌株的乙醇耐受性和发酵能力。结果 诱变菌株热带醋杆菌J-2276在不同体积分数乙醇胁迫下的生长状况均得到显著改善且能耐受体积分数为11%的乙醇,其发酵的麦芽醋中乙酸质量浓度较出发菌株热带醋杆菌J-22提高了65.0%。较高的乙醇耐受性也使诱变菌株热带醋杆菌J-2276产生更丰富的风味物质,其发酵的麦芽醋中总有机酸质量浓度和总挥发性物质质量浓度较出发菌株热带醋杆菌J-22均提高了45.3%。结论 该研究通过采用ARTP诱变和HTS相结合的方法选育出1株耐乙醇热带醋杆菌J-2276,且其连续传代7次后遗传性能稳定,有效提高了麦芽醋发酵的产酸量,改善了麦芽醋产品的滋味和风味,具有广泛的应用前景。

    Abstract:

    Objective This study aims to breed an elite strain of acetic acid bacteria with ethanol tolerance and suitable for malt vinegar brewing, thereby improving the utilization of raw materials, the yield of acetic acid, and the quality of the final vinegar product.Method The fermentation capacity of Acetobacter tropicalis J-22, Gluconacetobacter saccharivorans J-15, and Acetobacter pasteurianus HN 1.01 in beer with different volume fractions (6%~10%) of ethanol was compared. Based on the acetic acid yield (acetic acid mass concentration), ethanol conversion rate, and sensory evaluation, Acetobacter tropicalis J-22 , which achieved the highest overall assessment, was identified as the original strain. Acetobacter tropicalis J-2276 was finally bred through atmospheric and room temperature plasma (ARTP) mutagenesis combined with high-throughput screening (HTS). The growth curves and malt vinegar fermentation performance of the mutant strain Acetobacter tropicalis J-2276 and the original strain Acetobacter tropicalis J-22 under different volume fractions of ethanol further verified the ethanol tolerance and fermentation capacity of the mutant strain.Result The mutant strain Acetobacter tropicalis J-2276 exhibited significantly improved growth under ethanol stress at various volume fractions and tolerated the ethanol volume fraction up to 11%. Compared with the original strain Acetobacter tropicalis J-22, the mutant strain Acetobacter tropicalis J-2276 increased the acetic acid mass concentration of malt vinegar by 65.0%. Furthermore, the mutant strain Acetobacter tropicalis J-2276 with higher ethanol tolerance produced richer flavor compounds, with the mass concentrations of total organic acids and total volatile compounds increasing by 45.3% compared with the original strain Acetobacter tropicalis J-22.Conclusion This study demonstrates the successful breeding of an ethanol-tolerant strain, Acetobacter tropicalis J-2276, by ARTP mutagenesis and HTS. The strain exhibits stable genetic traits after seven successive generations, effectively increases the acid yield in malt vinegar fermentation, and improves the flavor quality of malt vinegar products, demonstrating broad application prospects.
