    【目的】建立一种简单、灵敏的金黄色葡萄球菌(Staphylococcus aureus)检测方法,用于食品中该致病菌的高效检测。【方法】以金黄色葡萄球菌为研究对象,基于指数扩增反应(exponential amplification reaction,EXPAR)结合DNA银纳米簇(P-AgNCs)开发了一种简单、灵敏的金黄色葡萄球菌荧光检测方法EXPAR-P-AgNCs。通过设计识别探针,可在金黄色葡萄球菌存在的情况下触发EXPAR,生成大量的靶标DNA(T),T与发夹P结合,富G序列远离P-AgNCs,从而降低P-AgNCs的相对荧光强度,实现了对金黄色葡萄球菌的检测。【结果】金黄色葡萄球菌浓度在1×102~1×106 CFU/mL时,相对荧光强度与其浓度呈线性相关,检测限达10 CFU/mL,且特异性明显,无交叉反应。在牛奶、猪肉样品中加入金黄色葡萄球菌,利用该方法测定金黄色葡萄球菌,回收率为94.5%~115.0%。【结论】所构建的检测方法灵敏度高、特异性好,为病原微生物及其他影响食品安全的污染物的检测提供了思路。

    Abstract:

    [Objective] A simple and sensitive detection method for Staphylococcus aureus is established for the efficient detection of this pathogen in food.[Method] Targeting Staphylococcus aureus, a simple and sensitive fluorescent detection method for Staphylococcus aureus, EXPAR-P-AgNCs, was developed based on exponential amplification reaction (EXPAR) combined with DNA silver nanoclusters (P-AgNCs). A recognition probe was designed to initiate EXPAR in the presence of Staphylococcus aureus, generating a large amount of target DNA(T). The binding of T to hairpin P displaced the G-rich strand from P-AgNCs, resulting in a decrease in the relative fluorescence intensity of P-AgNCs and enabling the detection of Staphylococcus aureus.[Result] The relative fluorescence intensity showed a good linear relationship with Staphylococcus aureus concentration in the range of 1×102 and 1×10? CFU/mL. The limit of detection was as low as 10 CFU/mL. The method shows high specificity without cross-reactions. The recovery rates for Staphylococcus aureus spiked in milk and pork samples ranged from 94.5% to 115.0%.[Conclusion] The developed detection method demonstrates high sensitivity and excellent specificity, offering a promising approach for the detection of microbial pathogens or other contaminants threatening food safety.
