    摘要:

    【目的】建立一种基于胶体碳的竞争抑制免疫层析法,用于检测奶粉和鸡肉中的麻保沙星(marbofloxacin,MBF)残留。【方法】用实验室自制的抗MBF多克隆抗体制备了胶体碳免疫探针,考察了反应体系的pH、重悬液、样品垫处理液、标准品稀释液、胶体碳质量浓度、一抗质量浓度、包被原质量浓度和二抗质量浓度对MBF胶体碳免疫层析法的影响,并测定了该方法的灵敏度、特异性、稳定性和准确性。【结果】该方法在最优条件下的消线值为4.0 ng/mL,较传统胶体金免疫层析法提高了5倍,半定量检测限为0.4 ng/mL;该方法对MBF检测表现出良好的特异性,可在室温下保存16周,在奶粉和鸡肉样品中的加标回收结果与超高效液相色谱荧光检测法基本一致。【结论】该方法操作简单、灵敏度高、特异性强且稳定性好,可用于动物性食品中MBF残留的快速检测。

    Abstract:

    [Objective] A colloidal carbon-labelled immunochromatographic assay with competitive inhibition was developed for the detection of marbofloxacin (MBF) residues in milk powder and chicken.[Method] The anti-MBF polyclonal antibody prepared in the author's laboratory was employed to produce the colloidal carbon-labelled immunoprobe. The effects of pH of reaction system, resuspension solution, treatment solution of sample pad, standard dilution buffer, mass concentration of colloidal carbon, mass concentration of primary antibody, mass concentration of coating antigen, and mass concentration of secondary antibody on the MBF colloidal carbon-labelled immunochromatographic assay were investigated. The sensitivity, specificity, stability, and accuracy of the method were determined.[Result] Under optimal conditions, the cut-off value of the method was 4.0 ng/mL,which was five times higher than the value of traditional colloidal gold-labelled immunochromatographic assay. The semi-quantitative detection limit was 0.4 ng/mL. The method showed good specificity for the detection of MBF and could be stored at room temperature for 16 weeks. The spiked recovery results in milk powder and chicken samples were basically consistent with those of ultra-high performance liquid chromatography coupled with a fluorescence detector.[Conclusion] The established method is simple, sensitive, specific, and stable, and thus it can be utilized for rapid detection of MBF residues in animal-derived foods.
