    【目的】 研究云南茶树花提取物(TFE)的体外抗氧化、抗炎活性及其对细胞衰老的抑制作用。 【方法】 通过紫外(UV)-可见分光光度法和高效液相色谱(HPLC)法分析测定TFE的主要活性成分,通过自由基清除试验测定TFE的体外抗氧化能力,利用UV诱导的人永生化表皮角质形成细胞(HaCaT)光老化模型测定TFE的抗炎活性。建立人成纤维细胞衰老模型,通过β-半乳糖苷酶(β-galactosidase,β-gal)染色法检测细胞衰老水平,通过酶联免疫吸附试验检测基质金属蛋白酶-1(matrix metalloproteinase-1,MMP-1)、基质金属蛋白酶-3(matrix metalloproteinase-3,MMP-3)和Ⅰ型前胶原蛋白(type I procollagen,PC-1)基因的表达。 【结果】 TFE中总黄酮、茶多酚、总蛋白质和总糖的质量分数分别为14.51%、12.83%、9.57%、50.34%,其中茶多酚以酯型儿茶素类化合物为主。DPPH、ABTS自由基清除能力的IC50分别为34.32、227.79 μg/mL,TFE显著抑制了UV诱导的HaCaT细胞中活性氧(reactive oxygen species,ROS)的生成,下调了IL-6TNF-α基因的表达,并上调了核因子E2相关因子2(nuclear factor erythroid-2 related factor 2,Nrf2)和血红素氧合酶-1(heme oxygenase-1,HO-1)基因的表达。此外,TFE还有效抑制了人成纤维细胞的衰老,下调了MMP-1MMP-3基因的表达,上调了PC-1基因的表达。 【结论】 TFE具有较强的体外抗氧化、抗炎活性,可通过抑制MMP-1MMP-3基因的表达,提高PC-1基因的表达,来延缓细胞衰老,具有潜在的抗衰老活性。

    Abstract:

    [Objective] This study aims to investigate the in vitro antioxidant and anti-inflammatory activities of Yunnan tea flower extract (TFE) and its Inhibitory effect on cell aging. [Method] The main active components in TFE were determined by ultraviolet (UV)-visible spectrophotometry and high performance liquid chromatography (HPLC). The antioxidant and anti-inflammatory activities of TFE were determined by the free radical scavenging assay and UV-induced photoaging model of human keratinocytes (HaCaT), respectively. After the aging model of fibroblasts was established, the aging level of cells was assessed by the β-galactosidase (β-gal) staining assay, and the expression of matrix metalloproteinase-1 (MMP-1), matrix metalloproteinase-3 (MMP-3), and type Ⅰ procollagen (PC-1) were determined by enzyme-linked immunosorbent assay. [Result] The mass fractions of total flavonoids, tea polyphenols, total proteins, and total sugars in TFE were 14.51%, 12.83%, 9.57%, and 50.34%, respectively. Ester-type catechins were the main tea polyphenols. The IC50 in scavenging DPPH and ABTS free radicals were 34.32 μg/mL and 227.79 μg/mL, respectively. TFE significantly inhibited reactive oxygen species (ROS) production-induced by UV in HaCaT cells, down-regulated the expression of IL-6 and TNF-α, up-regulated the expression of nuclear factor erythroid 2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1). In addition, TFE inhibited the aging of fibroblasts, down-regulated the expression of MMP-1 and MMP-3, and up-regulated the expression of PC-1. [Conclusion] TFE had strong antioxidant and anti-inflammatory activities. It can down-regulated the expression of MMP-1 and MMP-3 and up-regulated the expression of PC-1 to prevent cell aging, demonstrating potential anti-aging activity.
