    摘要:

    目的 通过考察益生菌发酵上清液对细胞光老化的改善作用,探究不同超滤组分能否作为有效成分应用于抗光老化的化妆品。方法 以鼠李糖乳酪杆菌发酵上清液(LR)、副干酪乳酪杆菌发酵上清液(LP1)和植物乳植杆菌发酵上清液(LP2)的3个不同超滤组分(相对分子质量>3 000、相对分子质量1 000~3 000和相对分子质量<1 000)为研究对象,构建紫外线(UVB)诱导的正常人皮肤成纤维细胞(NHDF)模型和小鼠黑色素瘤细胞B16F10模型,评价3种益生菌发酵上清液不同超滤组分的抗光老化作用。结果 LR、LP1和LP2的超滤组分均无细胞毒性,显著降低了SA-β-gal染色率、酪氨酸酶的酶活力、活性氧(ROS)水平、一氧化氮(NO)和肿瘤坏死因子-α(TNF-α)浓度,增加了ABTS+自由基清除能力,且相对分子质量<1 000的超滤组分作用最显著,其中酪氨酸酶活力相比于模型组降低50%~60%。结论 3种益生菌发酵上清液可以通过抑制黑色素生成、抗氧化和抗炎发挥抗光老化作用,相对分子质量<1 000的超滤组分可作为化妆品行业中潜在的抗光老化活性成分。

    Abstract:

    Objective The present study investigates the ameliorative effects of probiotic fermentation supernatants on cellular photoaging, exploring whether different ultrafiltration fractions can serve as effective components in anti-photoaging cosmetics.Method Three distinct ultrafiltration fractions (relative molecular mass >3 000, relative molecular mass 1 000~3 000, and relative molecular mass <1 000) of the fermentation supernatants of Lactobacillus rhamnosus (LR), Lactobacillus paracasei (LP1), and Lactobacillus plantarum (LP2) were examined for their anti-photoaging effects on UVB-induced human skin fibroblasts (NHDF) and mouse B16F10 melanoma cell models.Result All the ultrafiltration fractions of LR, LP1, and LP2 were non-toxic to cells and significantly reduced the SA-β-gal staining rate, tyrosinase activity, reactive oxygen species (ROS) level, and nitric oxide (NO) and tumor necrosis factor-α (TNF-α) concentrations, while increasing the ABTS+ free radical scavenging capacity. The ultrafiltration fraction with the relative molecular mass <1 000 showed the most pronounced effects, decreasing the tyrosinase activity by 50%~60% compared with the control group.Conclusion The fermentation supernatants of the three probiotics can exert anti-photoaging effects by inhibiting melanin production and exhibiting antioxidant and anti-inflammatory activities. The ultrafiltration fraction with the relative molecular mass <1 000 may serve as potential anti-photoaging active components in the cosmetic industry.
