    目的 探讨乳铁蛋白(Lactoferrin,LF)调制乳粉及其所含的α-乳白蛋白、N-乙酰神经氨酸、酵母-β葡聚糖和乳铁蛋白4种营养成分对RAW264.7巨噬细胞的免疫调节机制。方法 利用LPS和IL-4诱导M1/M2型巨噬细胞极化,并采用实时荧光定量PCR、流式细胞术和Western blot分析其对巨噬细胞极化及相关信号通路的影响。结果 乳铁蛋白调制乳粉可促进巨噬细胞吞噬活性,在10~50 μg/mL范围内剂量依赖性降低M1型极化并促进M2型极化。含4种营养成分的组合亦具类似作用,且优于3种成分组合。此外,乳铁蛋白调制乳粉可促进抗原呈递相关分子并提高抗氧化酶的活性,并通过抑制MAPK/NF-κB和增强Akt/STAT6通路调节M1/M2型巨噬细胞极化,改善炎症水平。结论 乳铁蛋白调制乳粉通过调控巨噬细胞极化来发挥免疫调节作用。

    Abstract:

    Objective This study aims to investigate the immunoregulatory mechanisms of lactoferrin formula and its four nutritional components (α-lactalbumin, N-acetylneuraminic acid, yeast β-glucan, and lactoferrin) in RAW264.7 macrophages.Method M1 and M2 macrophage polarization were induced by lipopolysaccharides (LPS) and interleukin (IL)-4, respectively. Quantitative real-time PCR, flow cytometry, and Western blot were employed to analyze macrophage polarization and related signaling pathways.Result Lactoferrin formula enhanced the phagocytic activity of macrophages and dose-dependently inhibited M1 polarization while promoting M2 polarization within the range of 10~50 μg/mL. A combination of four nutritional components exhibited similar effects, with greater effects than the three-component combination. Additionally, the lactoferrin formula upregulated antigen-presenting molecules and antioxidant enzyme activity. It modulated macrophage polarization by inhibiting the mitogen-activated protein kinase (MAPK)/nuclear factor (NF)-κB pathway and activating the protein kinase B (Akt)/signal transducer and activator of transcription 6 (STAT6) pathway, thereby alleviating inflammation.Conclusion Lactoferrin formula exerts immunoregulatory effects by modulating macrophage polarization.
