    建立了一种从酿酒酵母干细胞中高效抽提谷胱甘肽(GSH)的新方法,料液比为1:8,采用醋酸-盐酸调节抽提pH值为3.5,抽提温度(70±1)℃的条件下,GSH抽提量可达5.50 mg/g,比已报道的其它抽提法提高54.49%~62.24%。抽提液经微滤(膜孔径0.45、0.22μm)、超滤(膜截留相对分子质量5000)处理,蛋白质总去除率为60.98%。所得超滤液经高效液相法和氨基酸分析仪分析表明,粗提物中的杂质除蛋白质、小肽外,氨基酸类杂质主要为谷氨酸,为GSH的进一步精制提供了基础数据。

    Abstract:

    An efficient method for glutathione(GSH) extraction from the dried Sacchromyces cells was proposed in this study.During extraction at(70±1)℃,the mixture of acetic acid-hydrochloric acid was used to adjusted pH 3.5,the ratio of solid to liquid(w/v) was kept at 1:8. GSH extraction result reached at 5.5 mg/g,which was 54.49%-62.24%higher than that of all of the reported extraction process.After treatment through micro-filtration(0.45、0.22μm) and ultra-filtration(5 kD MWCO),60.98%of the yeast protein was removed.The collection from ultrafiltration was analyzed by high performance liquid chromatography and amino acid analyzer, the result showed that glutamic acid was the major portion in amino acids impurity besides protein and small peptide in crude extract,and these would provide basic data for purifying GSH.
