    为了明确蔗糖对七味白术散治疗菌群失调腹泻小鼠肠道细菌多样性的影响,将48只SPF级小鼠随机分成4 组,正常组小鼠灌胃无菌盐水,以头孢拉定、硫酸庆大霉素混合灌胃小鼠构建菌群失调腹泻模型,然后分别灌胃无菌盐水、七味白术散50%量超微汤药、七味白术散50%量超微汤药加糖,治疗结束后,提取肠道微生物宏基因组,通过扩增核糖体DNA限制性酶切片段分析(ARDRA)技术研究肠道细菌多样性,判断药效。结果表明,七味白术散50%量超微汤药组、七味白术散50%量超微汤药加糖组及正常组的条带数和OTU数均为6,多样性指数分别为2.322、2.585、2.585;相似系数分别为0.363 6、0.277 8、1.000 0;从相似系数分析发现,七味白术散50%量超微汤药加糖组相似系数低于七味白术散50%量超微汤药组;聚类分析、主成分分析表明,七味白术散50%量超微汤药组较七味白术散50%量超微汤药加糖组更接近正常组,说明七味白术散50%量超微汤药组与七味白术散50%量超微汤药加糖组均能治疗小鼠菌群失调腹泻,且七味白术散50%量超微汤药组对小鼠的治疗效果优于七味白术散50%量超微汤药加糖组。七味白术散在临床用药时不加蔗糖更理想。

    Abstract:

    In order to evaluate the effect of sucrose on Qiweibaizhusan to intestinal bacteria diversity in dysbacteriotic diarrhea mice,48 SPF mice were randomized into 4 groups. Normal control group of mice were via intragastric administration of sterile water. Model group and treated groupmice were constructed via intragastric administration of cefradine and gentamycin sulfate. Model group mice were via intragastric administration of sterile water,and treatment groups were via intragastric administration of 50% amount of ultra- micro Qiweibaizhusan powder and 50% amount of ultra-micro Qiweibaizhusan powder with 8% sugar respectively. After treatment,intestinal microbiotia metagenome DNA were extracted. The intestinal bacteria diversity was analyzed by amplified ribosomal DNA restriction analysis. The results showed that OTU and stripe numbers in 50% amount of ultra-micro powder Qiweibaizhusan group,50% amount of ultra-micro powder Qiweibaizhusan with sucrose group and normal control group were all 6. The diversity index in 50% amount of ultra-micro powder Qiweibaizhusan group,50% amount of ultra-micro powder Qiweibaizhusan with sucrose group and normal control group were respectively 2.322,2.585 and 2.585. The same similarity coefficient in 50% amount of ultra-micro powder Qiweibaizhusan group,50% amount of ultra-micro powder Qiweibaizhusan with sucrose group and normal control group were respectively 0.363 6,0.277 8 and 1.0000. The similarity coefficient in 50% amount of ultra-micro powder Qiweibaizhusan with sucrose group is lower than that in 50% amount of ultra-micro powder Qiweibaizhusan group. Cluster analysis and principal component analysis showed that 50% amount of ultra-micro powder Qiweibaizhusan group were better than 50% amount of ultra-micro powder Qiweibaizhusan with sucrose group to normal control group. Both 50% amount of ultra-micro powder Qiweibaizhusan and 50% amount of ultra-micro powder Qiweibaizhusan with sucrose could cure dysbacteriosis diarrhea. The former had better effect than the latter. It was advised to adopt the ultra-micro powder Qiweibaizhusan without the additive of sucrose.
