    Abstract:

In order to obtain high enzyme activity of β-galactosidase,twenty-eight strains which produced β-galactosidase were screened from soil using lactose as inducer and X-gal as indicator.Four strains which have higher hydrolysis enzyme activity were screened by shake flask culturing. One strain B5582Y which have both of hydrolysis and transgalactosylation Activity was screened by HPLC,its hydrolysis of initial enzyme activity can reach upwards of 12 U/mL. And it was identified as Klebsiella michiganens is according to the cell morphology, physiological and biochemical characteristics,16SrDNA and rpoB functional gene sequence.
