    Abstract:

    In order to isolate high quality DNA from Wheat Qu the effect of DNA isolation by seven different methods were evaluated. The extracted DNA was analysised by gel electrophoresis,PCR,ultraviolet spectrophotometry,and real-time fluorescent PCR,respectively. The results showed that SDS-CTAB method has the best effect on the isolation of total DNA of Wheat Qu. This method' products showed less contamination from protein and polysaccharide,the yield of the DNA was reached 149.6 ng/μL and its' relative account of template is almost 2.343 and 1.753 times in bacteria and fungi respectively compared to SDS method.
