    Abstract:

    To explore the possible sequence and function of internal ribosme entry site(IRES) in flanking sequence of FOSB. Using the typical bicistronic reporter system of dual-luciferase,constructing the reporter plasmids containing the 5'UTR of FOSB mRNA and then transfecting them into HEK293 cell and 4 strains of cancer cells. Performing the deletion of FOSB mRNA 5'UTR and stimulating the Bel-7402 cell with anticancer drug,paclitaxel or cis-platinum. The results showed that the FOSB mRNA 5'UTR has IRES activity and the sequences of 160nt between -376 and -217 nt are essential to its IRES function. The FOSB IRES activity can be stimulated in Bel-7402 treated with paclitaxel or cis-platinum,especially under the stimulating of paclitaxel. All these findings provide a new thought and direction for the study of FOSB in cancer prevention and treatment.
