Abstract:在反应初期,壳聚糖的脱乙酰度随温度的升高而增大,而后反应速率减慢,脱乙酰度的变化也减小。当NaOH溶液质量分数达到45%左右、温度达100℃以上后,反应速率急剧提高,脱乙酰度也发生较大变化。对于NaOH溶液的加热,与常规加热方式相比,微波具有快速加热的优越特性。利用H-NMR研究发现,微波对甲壳素脱乙酰反应有一定的“非热效应”。

Abstract:以月桂酸甲酯和β-氨基丙腈为原料,在有机相中经脂肪酶催化合成N-月桂酰-β-氨基丙腈(LAP)。在比较所用脂肪酶的催化活性的基础上,详细探讨了固定化脂肪酶Candida antarctica(南极洲假丝酵母)CAL-2催化该反应的影响因素。经筛选,适宜的反应条件为:70℃,n(β-氨基丙腈):n(月桂酸甲酯)=1:1,底物质量浓度为35g/L,加酶量为200U/g(以氨基丙腈的质量计),反应体系初始加水量为3%腈重。在此实验条件下反应24h后,β-氨基丙腈的最高转化率为96.8%。

Abstract:通过流化床干燥法制备稳定化颗粒酶,提高酶的耐热性。在制备过程中进风温度为40℃,酶活回收率达到90%以上。采用海藻酸钠(SA)作包被剂,随着SA质量浓度的提高,酶的耐干热性先增加后降低,在0.4g/dL最高,而其耐湿热性不断增强。水分活度提高,固态酶的耐热性就随之降低,在Aw≥0.3时流化床干燥酶样品的耐热性都明显高于原酶粉,且SA质量浓度越高,酶对水分的耐受能力越高。在流化床干燥酶制剂中添加硫酸铵、氯化钠、硫酸钠等盐类稳定剂,酶的耐干热性有的增强,有的减弱,而其耐湿热性都有所增强,其中硫酸铵的热稳定化效果最好。酶在高温下的失活速度遵循一级动力学,即它的失活是由酶蛋白变性引起的,流化床干燥样品的失活常数低于原酶粉的失活常数,而不同温度下酶的失活遵循阿累尼乌斯方程。

Abstract:研究了不同的加热温度、加热时间和加热方式对8种西非鱼糜冻藏后凝胶特性的影响。结果显示这些鱼糜基本上在40℃加热时具有较强的凝胶强度,60-70℃为凝胶劣化温度段;采用40℃加热20min后再在90℃加热40min的二段加热方式,可使鱼糜制品具有较高的硬度、弹性和凝胶强度。此外,马鲛鱼、竹英鱼和花鲷具有较强的抗蛋白质冷冻变性的能力,并具有较高的凝胶强度。

Abstract:L68菌株中的邻苯二酚 2 ,3 双加氧酶经硫酸铵分级沉淀、DEAE SepharoseFastFlow柱层析、Hydroxyapatite柱层析、Sephadex G15 0凝胶过滤分离 ,并经SDS 聚丙烯酰胺凝胶电泳检测 ,达到了电泳纯 .提纯倍数为 2 34 .1倍 ,收率为 2 0 .2 % .酶相对分子质量为 (132± 10 )kDa ,由 4个相对分子质量相同的亚基组成 .最适反应温度 35℃ ,且酶活力在 15~ 65℃比较稳定 ;最适 pH为 8.0 ,在 pH 7.0~ 10 .5酶活力较高且比较稳定 .某些金属离子尤其是Fe3+ 对酶活力有强烈的抑制作用 .以邻苯二酚为底物 ,Km 为 2 0 .63μmol/L ,Vmax为 2 .82 μmol/ (min·mg)

Abstract:将重组过氧化氢酶和市售一种商品过氧化氢酶的性质进行了比较。重组酶的最适温度为70℃,某商品过氧化氢酶的最适温度为40℃;与这种商品过氧化氢酶相比,重组酶的热稳定性更好。重组过氧化氢酶和这种商品过氧化氢酶的最适pH都是7.0;重组酶和这种商品过氧化氢酶在pH3-8的范围内均较为稳定;重组酶在室温放置1个月后酶活力基本保持不变,而此种商品过氧化氢酶活力损失约70%;金属离子对重组酶和这种商品过氧化氢酶都有抑制作用,抑制程度有所不同;EDTA抑制这种商品过氧化氢酶的活性,但不抑制重组酶的活性。

Abstract:采用正交试验设计,探讨了蛋白酶种类、腌制时间、腌制温度对风鸭风味和游离氨基酸产生,及亚硝酸钠、异抗坏血酸钠、乳酸用量和腌制时间对风鸭发色的影响。结果表明,蛋白酶种类、腌制时间、腌制温度对风鸭风味及游离氨基酸产生,及亚硝酸钠、异抗坏血酸钠、乳酸用量和腌制时间对风鸭的发色,均有显著性的影响。其最佳腌制条件:蛋白酶种类为毛霉蛋白酶 青霉蛋白酶 木瓜蛋白酶,腌制时间5h,腌制温度18℃,亚销酸钠、异抗坏血酸钠和乳酸用量分别为100mg/kg、300mg/kg和250mg/kg。
