Abstract:将大连蛇岛蝮蛇毒类凝血酶(Gloshedobin)基因克隆到载体pET32-a(+)上,在T7lac启动子下以N端融合硫氧还蛋白的形式在大肠杆菌BL21(DE3)中进行表达,诱导条件为:30℃诱导6 h,IPTG浓度为1 mmol/L.利用固定化金属离子(Ni2+)亲和层析分别对胞内可溶物和变性条件下的包涵体进行纯化.通过SDS-PAGE检测融合蛋白的纯度,采用点杂交和纤维蛋白凝结活性检测分析,显示纯化产物具有较高生物活性.

Abstract:用Brevibacterium sp.DGCDC-82在7L发酵罐中分批培养.分别对添加剂吐温-80、培养温度、培养基的pH、通风量和搅拌速度在胆固醇氧化酶的生产中的影响进行了研究.结果显示以上条件均对产酶有影响.在不同的发酵阶段改变发酵操作条件,发酵20 h最高酶活可达1203U/L,生产强度可达60 U/(L·h).既有效地提高了胆固醇氧化酶的产量,又防止了发酵过程中的泡沫外溢.

Abstract:对准纳米银对番茄汁、番茄-胡萝卜混合汁等蔬菜汁的防腐功效及准纳米银与其它防腐剂的复合防腐效果进行了研究.结果表明,当准纳米银溶液单独用于番茄汁作为抗菌防腐剂时,其有效质量浓度为0.1 mg/L;当准纳米银溶液单独用于番茄-胡萝卜混合汁防腐时,0.1 mg/L的质量浓度同样可以起到较好的保鲜效果,但是必须给予适度的热力杀菌;当番茄汁采用准纳米银-尼泊金乙酯复合防腐剂处理时,其最佳组合为:准纳米银0.04 mg/L,尼泊金乙酯200mg/L;而这种复合处理用于番茄-胡萝卜混合汁时,则需要在95~100℃条件下杀菌5 min.
