Abstract:The damages of endothelial cells may cause varieties of vascular diseases. In order to protect endothelial cells against damageandimprove functions ofendothelial cells,we can make use of botanical active substances to act on these cells. The paper summarizes the classifications,sources and functions of botanical active substances that have functions of protecting endothelial cells from damages. It also lists the common models of endothelial cell damage,and compares the situation of studies on the protection of botanical active substances to the endothelial cell damages. The paper tries to provide theoretical foundation for the large-scale using of botanical active substances in the future,expecting it to be a trend to use the botanical active substances on the damages of endothelial cells. It aims to prevent and treat vascular diseases.

Abstract:To explore the anti-tumor effect of three cervical cancer cell lines Hela,Siha and C-33A induced by gossypol acetic acid(GAA) respectively and the mechanism. The inhibitory effect of GAA on the growth of three cervical cancer cell lines was detected by MTT assay.The DNA damage of three cervical cancer cell lines caused by GAA was detected by single cell gel electrophoresis. The expression of phosphorylated histone protein(γH2AX) in three cervical cancer cell lines produced by GAA was detected by immunohistochemistry. Results:GAA(0~160 μmol/L) inhibited the growth of three cervical cancer cell lines in a dose-time dependence. The inhibition rate increased with the increase of concentration and time. 2.5~40 μmol/L of the GAA for 24 h,or 20 μmol/L GAA role 3~48 h,The result of single cell gel electrophoresis display that cell comet head DNA percentage was reduced,but tail length,comet length,tail DNA percentage,tail moment and olive tail moment was increased;and immunohistochemistry results showed that,the γH2AX positive rate increased in a concentration-time dependence. The mechanism of antitumor by GAA is to inhibit the proliferation of three cervical cancer cell lines,induce DNA double strand breaks,and lead to cell death. And the sensitivity of C-33A was the highest,the Hela was the second,and Siha was the most insensitive.

Abstract:To investigate the effect of nutrients on furan formation from glucose/galactose model systems,different doses of minerals(iron,zinc,and magnesium),unsaturated fatty acids(linoleic acid,linolenic acid) and ascorbic acid were added to these model systems. After heated in oil bath at 121 ℃ for 30 min,these model systems were analysed by headspace gas chromatography-mass spectrometry(HS-GC-MS). The results revealed that the generation of furan from both model systems increased rapidly(p<0.05) when the content of iron was elevated. High concentration of zinc(≥10 mg) and magnesium have obvious inhibitory effect on furan formation from glucose model systems(p<0.05);while a relatively high concentration of zinc(≥5 mg) and high concentration of magnesium(>10 mg) could inhibit the formation of furan from galactose model systems(p<0.05). With the addition of unsaturated fatty acids,the amount of furan has obviously increased for both glucose and galactose model systems(p<0.05). Ascorbic acid has obvious promoting effect on furan formation from glucose model,but galactose model was the opposite(p<0.05). Thus,this study could be regarded as a theoretical reference for optimizing processing conditions and inhibiting the formation of furan in heat processed food.

Abstract:Epoxide hydrolase can be effectively used in the preparation of optically active epoxides and vicinal diols,which are versatile intermediates for the preparation of highly value-added bioactive compounds such as pharmaceuticals. In this study,a gene encoding a novel epoxide hydrolase(VrEH3) was successfully cloned from Vigna radiata by RT-PCR and T-hairpin structure-mediated PCR amplification(THSO-PCR) techniques. The cloned coding region DNA sequence of Vreh3 is 1 178 bp in length,harboring two introns with sizes of 142 bp and 79 bp and a 957 bp open reading frame that encodes a 318-aa VrEH3. The theoretical molecular weight and calculated pI of VrEH3 were 36.2×103 and 5.59,respectively. The catalytic active center was constituted by a strictly conserved catalytic triad of Asp101-Asp262-His297. Subsequently,the expression plasmid pET-28a(+)-Vreh3 was constructed by ligated Vreh3 with pET-28a(+) and transformed into Escherichia coli BL21(DE3),forming a recombination strain E. coli/Vreh3. Using the E. coli/Vreh3 as the catalysis,the(R)-1-phenyl-1,2-ethanediol was obtained with a yield of 79.4% and an enantiomeric excess value of 94.7% by enantioconvergent hydrolysis of(R,S)-styrene oxide.

Abstract:To build a method that Curcumin(Cur) of Curcumin novel nano emulsion,CNNE) were measured by HPLC,study the pharmacokinetic characteristics of the CNNE,and compare the bioequiavailability of CNNE and Cur. CNNE was prepared by water droplets method,and plasma drug concentrations of different time points were measured by HPLC after oral administration CNNE and Cur. The curve of AUC was drew by excel software,and the main pharmacokinetic parameters and bioequiavailability were calculated by DAS2.1.1 software. The parameters of compartmental model,AUC(0-∞) of CNNE and Cur,were(3 439.27±180.63) (μg·h)/L,(482.07±42.14) (μg·h)/L,and the parameters of compartmental model,AUC(0-∞) of CNNE and Cur,were(3 410.20±154.09) (μg·h)/L,(446.66±44.02) (μg·h)/L. The relative bioavailability of compartmental model and non-compartmental of CNNE are 713.44%,763.49%,respectively. The results show that prepared CNNE promoted absorption of Cur,and the bioavailability of CNNE is about seven times as much as Cur. CNNE and Cur are not bioequivalent.

Abstract:The dielectric properties of taro slices with different moisture content from 25% to 85% at 20,40,60 ℃ and 80 ℃ are measured by the open-ended coaxial-line probe and a network analyzer from 30 to 3 000 MHz.The results show that dielectric constant and dielectric loss factor decrease with the increase of frequency,the dielectric constant increase with the increase of moisture content.the dielectric constant and dielectric loss factor of taro decrease with the increase of temperature.Under the condition of 915 MHz,the mathematical model's levels of significance of dielectric constant and dielectric loss factor are both less than 0.000 1,this result can predict the moisture content of the taro.the taro intake depth of the same moisture content and temperature decreases with the increase frequency,the taro intake depth increases with the increase of temperature under a certain frequency.

Abstract:In order to investigate the quality characteristics of different cuts of yak meat,the L* value,a* value,b* value,press loss,cooking loss,shearing force were determined,and the distribution of quality traits of yak meat among different cuts was analyzed using by analysis of variance,principal component analysis and hierarchical cluster analysis. There were significant differences in quality traits among various cuts(P<0.01). Three principal component factors could be exacted with PC1 of 35.11%,PC2 of 25.13%and PC3 of 16.63% through principal component analysis. It could distinguish cuts of tenderloin,chuck tender,foreshin,hindshin,ribeye. Hierarchical cluster analysis classified 17 varieties into two main groups on the basis of the measured parameters,this closely related to the water holding capacity and tenderness. The result showed that there were significant differences in quality traits among various cuts and the yak meat quality from the rear part of the body better than the front part.

Abstract:In this study,barley seedlings were used as main materials to produce barley ferment,which was fermented by yeast. The single factor experiment method was applied for research on the fermentation conditions. The optimum processing parameters are as follows:32 ℃ of fermentation temperature,0.67% of yeast inoculated,5.3% of sucrose added,and 12h of fermentation time. Preliminary study on antioxidant properties of this product was also conducted. The results show that barley ferment has a high antioxidant capacity.

Abstract:The effect of oat beta- glucan and protein on oat starch digestion and related mechanism were studied in this paper. Methods:Oats grain was processed into flour and then,sieved into whole oat flour samples with different particle sizes which contain different amounts of β-glucan(O-glu) and protein,and an in vitro starch digestion assay revealed different starch digestibility among those flour samples. RVA analysis also showed different profiles,and the addition of β-glucan did not significantly affect the starch digestion while pretreatment with protease increased the rate of starch digestion. Confocal microscopy analysis showed some complexed structure formed among β-glucan,protein and starch. Thus,it seems that the effect of β-glucan on starch digestion needs certain structural forms that are natively present in oat flour,and a matrix structure formed by linking with other components such as protein,might be the basis for slow digestion property of starch in the whole oat flour.

Abstract:In order to avoid the bad effect on enzyme catalytic efficiency and reuse with traditional insoluble carrier,a new kind of smart polymer named Eudragit S-100 was used for the immobilization of porcine pancreas lipase(PPL). Eudragit S-100 is an anionic copolymer of methacrylic acid and methylmethacrylate,and has a solubility reversible. The results showed that the modification rates of free amino of PPL immobilized with Eudragit S-100 reached 52.4% at 15 ℃ after 6 hours. The fluorescence emission spectra indicated that the enzyme molecule peptide chains were fold,and finally the structure became compact. The optimum temperature of immobilized PPL did not change,but the optimum pH moved slightly to an alkaline direction. Eudragit S-100 immobilization enhanced the enzyme stability against changes of pH and temperature. Furthermore,the immobilized PPL gained the solubility reversible on the basis of pH,and they had a high reusability,retaining about 45% of its original activity after five cycles of repeated uses.

Abstract:A method for the simultaneous determination of 12 persistent organic pollutants(POPs) for example hexachlorobenzene,α-666,heptachlor,etc in highly active dry yeast powder was developed using accelerated solvent extraction(ASE)-QuEChERS-gas chromatography-mass. The Sample was extracted by acetonitrile with accelerated solvent extraction apparatus,and then cleaned up by PSA. The qualification and quantification of target compound were performed by gas chromatography-mass in selected ion monitoring mode(GC-MS-SIM).The results show that the 12 POPs were linear in the range of 0.025~2.500 μg/mL with good correlation coefficients(R2≥0.992). The average recoveries of the 12 POPs in highly active dry yeast powder were 74.5%~ 111.7 % with relative standard deviations(RSDs,n=6) of 3.1%~11.3%,at the spiked level of 0.05,0.25,1.00 mg/kg. The limits of detection of the method(LODs)were 0.002~0.009 mg/kg. The method is convenient operation,rapid,saving on solvent ,could meet determination of 12 POPs residual in highly active dry yeast powder.

Abstract:To investigate whether the extract with ethanol from Anthemis nobilis L. could be ascribed to the inhibition of tyrosinase and know about the active ingredients,the experiment was conducted. The inhibition rates of different concentration of ethanol elution on mushroom tyrosinase were tested by the colorimetric method using L-tyrosine as substrate. It finds out that the 30% volume concentration of ethanol elution content is the most sensitive part on tyrosinase activity inhibition. The further result shows that the active part include kaempferol-3-O-β-D-glucopyranoside,isorhamnetin-3-O-β-D-glucopyranoside,kaempferol-3-O-(6"-O-methyl)-β-D-glucopyranoside.

Abstract:Polycaprolactone(PCL) and ceramsite were used as the solid denitrification carbon source and biofilm carrier respectively for the efficiency enhancement of denitrification. The performance of dissolved organic carbon(DOC) releasing of PCL as well as its roles in the denitrification process was investigated under microbial and amicrobic condition. Further research about the effect of ceramsite addition on denitrification efficiency was carried out by blending PCL with different dosages of ceramsite. Results showed that the DOC releasing rate was fast in the initial period(24 h) with the value of 0.656 mg/(g·d),then became slow and stable(0.095 mg/(g·d)),under amicrobic condition. However,the DOC releasing rate under microbial condition(0.286 mg/(g·d)) was much faster than those under amicrobic condition(0.123 mg/(g·d)),manifesting a 1.33 times of enhancements. This indicated that biological processes could significantly increase the efficiency of DOC release from PCL. Moreover,addition of ceramsite to PCL could improve the denitrification performance. The addition of 20,50 g and 100 g ceramsite to 100 g PCL led to the increase of denitrification rates from 0.813 mg/(L·h)(no addition) to 0.894 mg/(L·h),1.015 mg/(L·h) and 1.056 mg/(L·h),respectively. In addition,no NO2--N accumulation was observed during these processes,while a bit of soluble microbial products(SMP) accumulation was identified by Fluorescence EEM spectra.

Abstract:To establish a method for the total coumarins content at the wavelength of 348 nm,scopoletin was used as the reference substance.The linear range was 0~0.020 mg/mL,r=0.999 2. The average recovery was 102.44%,RSD=1.81%(n=9). Ultraviolet spectrometry was developed to determine total coumarins in noni juice fermented with different time. The contents of Noni jucie fermented with 25~500 d were in 0.176~0.243 mg/mL range. This method is simple,rapid,accurate and repeatability,which can be applicable for determination of coumarins content in liquid samples and the result was stable with the fermentation time extended.

Abstract:Two stains of Aspergillus oryzae were selected in combinated of koji making,in order to shorten koji making time and increase activity of neutral protease and aminopeptidase. The two strains of Aspergillus oryzae were combinated by appropriate proportion during koji making. The activity of neutral protease and aminopeptidase were determined. Single strain koji was cultivated for pH 7.2,30 ℃. The neutral protease activity of CICC2339 and CICC2066 were reached 1 187.7 U/g and 830.8 U/g on 42 h respectively,the aminopeptidase were reached 93.2 U/mL and 201.4 U/mL on 42 h and 60 h respectively. The highest activity of neutral protease and aminopeptidase of optimal combination A6B4 were reached 1 366.8 U/g and 148.7 U/mL on 42 h respectively,which had advantages than single strain. The combination could shorten koji making time,increase the activity of neutral protease and aminopeptidase. These results provided some assistance to the further understanding of Aspergillus oryzae in the food fermentation.

Abstract:Comparing decolorization rate and polysaccharide retention rate could be used as reference indexes,throughadopting the static adsorption to assess the best macroporous resins among AB-8,LSA700,LSD762,D101 and D900.The result showed D900 macroporous resin was the best while decolorizing polysaccharides. Besides,on the basis of Box-Behnken design principles,we adopted the method of response surface methodology in order to optimizing the resin decolorization technology of Dendrobium nobile Lindlpolysaccharides. The results turned out that thedecolorization temperature was superior to the time and the dosage of resin.Simultaneously,the interaction effects of time with temperature and resin on the decolorization rate had significant influence.The optimal decolorization conditions were as follows:decolorization time of 3.7 h,temperature of 40 ℃ and 0.33 g/mL resin. Under this conditions,the actual decolorization rate and polysaccharide retention rate of Dendrobium nobile Lindlpolysaccharides were 82.7 % and 76.5 %.

Abstract:In order to isolate high quality DNA from Wheat Qu the effect of DNA isolation by seven different methods were evaluated. The extracted DNA was analysised by gel electrophoresis,PCR,ultraviolet spectrophotometry,and real-time fluorescent PCR,respectively. The results showed that SDS-CTAB method has the best effect on the isolation of total DNA of Wheat Qu. This method' products showed less contamination from protein and polysaccharide,the yield of the DNA was reached 149.6 ng/μL and its' relative account of template is almost 2.343 and 1.753 times in bacteria and fungi respectively compared to SDS method.
